‘Tiramisu’ mengisahkan cinta antara dua insan yang sudah berkawan rapat di zaman sekolah, Melur (Sharifah Amani) dan Dani Mirza (Pierre Andre). Walaupun Dani seorang pelajar lelaki yang popular dan diminati oleh ramai pelajar perempuan tetapi Melur yang baru berpindah ke sekolah tersebut tidak meminati Dani seperti pelajar lain. Ini membuatkan Dani ingin mendampingi Melur dan berkenalan dengannya. Sebenarnya, Melur mempunyai masalah keluarga kerana abang iparnya seorang kaki judi. Malah, Melur sering diganggu oleh pemiutangnya dan Dani telah menyelamatkannya. Sejak itu, Melur dan Dani mula berkawan. Terdapat keserasian antara Melur dan Dani tetapi mereka tidak memikirkan soal cinta kerana usia masih muda.
Dani menyimpan cita-cita untuk menjadi seorang Chef Pastry walaupun ibunya, Puan Gai (Sheila Mambo) mengharapkannya menjadi seorang peguam yang berjaya. Melur sentiasa memberi sokongan kepada Dani dan mencadangkannya supaya memujuk ibunya dengan membuatkan kek Tiramisu. Akhirnya, Puan Gai mengalah dan menyokong cita-cita Dani.
Sebelum Dani menyambung pelajaran ke London, Dani berjanji sekiranya mereka masih bujang pada usia 28 tahun, dia mahu mengahwini Melur. Melur terharu. Mula-mula, mereka masih berhubung melalui emel. Lama-kelamaan, mereka terputus hubungan dan membawa haluan kehidupan masing-masing.
Beberapa tahun kemudian, Dani telah kembali ke tanah air sebagai Chef Pastry terkenal di London. Dani bercadang untuk membina empayar di Malaysia. Selain itu, Dani juga mahu mencari Melur namun kecewa kerana Melur telah berpindah. Lyana (Tania Hudson) yang meminati Dani sejak zaman persekolahan masih terpikat dengannya dan menaruh harapan supaya Dani akan jatuh cinta dan mengahwininya suatu hari nanti.
Dani dan Melur pernah berselisih beberapa kali tetapi tidak pernah bertembung muka. Suatu hari, Dani terserempak Melur bersama seorang budak perempuan, Ayesha (Mia Sara Nasuha). Dia berasa kecewa kerana menyangka Melur telah berkahwin. Sebenarnya, Melur berkorban untuk keluarganya selepas kakaknya bercerai dan bekerja di Singapura.
Sejak itu, Melur bekerja di kedai bunga dan menjaga anak saudaranya, Ayesha. Walaupun Dani dapat tahu bahawa Melur masih bujang tetapi Faris (Zain Ruffedge) tidak pernah berputus asa untuk memikat hati Melur telah mengaku dirinya sebagai teman lelaki Melur. Sekali lagi, Dani memilih untuk mengundur diri.
Masih adakah jodoh untuk Melur dan Dani? Apakah yang akan terjadi kepada mereka?
